Synergistic activity of ¥â-lactam antibiotics in combination with rifampin against Staphylococcus aureus isolates

Abstract
Thirty-two strains of Staphylococcus aurous isolated from various clinical samples were submitted to the combined antibiotic activity test of rifampin (RF) with ¥â-lactam antibiotics.
Preliminary drug-susceptibility test showed that the minimum inhibitory concentration (MIC) distribution patterns of the tested (3-lactams were more or less the same with those of RF except for ampicillin (AP), to which a considerable number of strains susceptible to all the other tested antibiotics, methicillin (MT), carbenicillin (CB), moxalactam (MX), and cefoperazone (CF), exhibited resistance while no AP-susceptible strain was detected among RF-resistant ones.
A simplified modification, in which fixed dilution of RF, one-fourth MIC, was mixed with the serial two-fold dilutions of the other drug (AP, MT, or CB), was mainly employed in the present combined antibiotic activity test instead of the conventinal checkerboard dilution and isobologram technique, which was, used occasionalyy only for reference. The results obtained revealed no significant difference between the two procedures.
RF turned out to have a marked enhancing antibacterial activity when used in combination with ¥â-lactam. RF-AP combination had synergistic effect on 26
strains among the total tested ones (81.2%). RF-MT had the similar effect on 17 strains (53.2%) and so did RF.CB on 13 ones (40.6%), whereas antagonistic action was observed only in the range of 6.2~15.6% throughout the test. The degree of individual drug resistance seemed to be related with the efficiency of the antibiotics combined, i.e., the more resistant to each antibiotic, the more susceptible were cells to the antibiotics combined.
